What is sirolimus?

Sirolimus is a drug that lowers the action of your body's immune system. The immune system tends to fight the presence of a transplanted organ such as a liver or kidney following surgery. Sirolimus is one element of a combination treatment, along with cyclosporine and steroids, to prevent the rejection of a transplanted kidney. In combination with other medications, sirolimus can also help treat a rare lung disorder called lymphangioleiomyomatosis.

How do I take sirolimus?

Take sirolimus orally on an empty stomach. Patients can take this medication with or without food, but it must be taken consistently using the chosen method. Swallow this medicine whole without breaking, crushing, or chewing it before swallowing. If you are unable to swallow sirolimus whole, talk to your doctor about alternative delivery methods. Be sure to wash hands thoroughly immediately after taking the drug sirolimus.

Before taking sirolimus, tell your doctor if:

  • You are pregnant, planning to become pregnant, or nursing
  • You are taking any prescription or non-prescription medicine
  • You have allergies to medicines, foods, or other substances
  • You have liver problems, kidney problems, a history of lung or breathing problems, high cholesterol or triglycerides, diabetes or high blood sugar, or low blood platelet levels
  • You are on dialysis
  • You have recently received or are scheduled to receive a vaccine
  • You have a history of tuberculosis (TB), or have ever had a positive TB skin test
  • You have had multiple organ transplants
  • You have had a failed organ transplant
  • You have a family history of skin cancer
  • You previously took cyclosporine and have recently stopped taking it

What are the possible side effects of taking sirolimus?

Patients may encounter some common side effects when taking sirolimus such as headache, constipation, diarrhea, joint pain, nausea, or stomach pain. Severe side effects may include calf or leg pain, redness, tenderness, or swelling; chest pain; fainting; fast or irregular heartbeat; menstrual changes; muscle pain, cramps, or tenderness; new or worsening breathing problem or cough; night sweats; red or blistered skin; severe or persistent headache; skin growths; swelling of the hands, ankles, feet, stomach, mouth or tongue; high blood sugar; infection; kidney problems; liver problems; unusual bleeding or bruising; lumps; tiredness; weakness; weight change;, vision changes; and wound healing problems.

What if I forget to take a dose of sirolimus?

Take a missed dose of sirolimus as soon you as you realize that you forgot one. If it is almost the time that you normally take your next dose, skip your missed dose and return to your regular dosing schedule. Never take a double dose of medication to make up for missed doses.

How do I store sirolimus?

Keep sirolimus at room temperature, between 68 and 77 degrees Fahrenheit (20 and 25 degrees Celsius). Store this medication in a tightly closed container, and keep the container away from any heat, moisture, or light. Keep drugs in their outer container even if they come with an additional blister container. Do not use any expired medicine. Store all medications out of reach of children and pets.

What happens if I overdose on sirolimus?

Call the Poison Control Center 1-800-222-1222 or get help from either your local poison control center or the nearest emergency room immediately.

Warnings

Sirolimus is likely to lower your body's ability to fight infection. When possible, avoid contact with people who have diseases or infections. Inform a doctor immediately if you begin to notice signs of an infection such as fever, sore throat, chills, or rash. Infections may become severe and, rarely, fatal. Use of this medication may also cause an increased risk of certain types of cancer, including lymphoma and skin cancer.
